چکیده

PURPOSE. The ocular surface epithelium is part of the mucosal defense system. Because transcription factor NFB in mucosal epithelial cells plays a central role in regulating the genes that govern the onset of mucosal inflammatory responses, we examined the role of a regulator of NFB, I B , in murine ocular surface inflammation. METHODS. The eyes of I B / mice were analyzed biomicroscopically and histologically. I B expression in normal mouse cornea and conjunctiva was examined by RT-PCR. The results were compared with those obtained in other tissues by realtime PCR. I B mRNA on the ocular surface and in other mucosal tissues was localized by in situ hybridization. RESULTS. I B / mice manifested chronic inflammation, specifically in the ocular surface, but not in other tissues. In normal mice, I B was expressed in a variety of mucosal tissues. The I B transcript was predominantly distributed in the epithelia of these tissues. As inflammatory symptoms progressed on the ocular surface of I B / mice, inflammatory cells, mainly CD45R/B220 and CD4 cells, intensely infiltrated the submucosa of the conjunctival epithelia. This infiltration was accompanied by an almost complete loss of goblet cells in the conjunctival epithelia.

Modulation of Conjunctival Goblet Cell Function by Inflammatory Cytokines

Ocular surface inflammation associated with Sjögren's syndrome is characterized by a loss of secretory function and alteration in numbers of mucin secreting goblet cells. Such changes are a prominent feature of ocular surface inflammatory diseases and are attributed to inflammation; however, the exact effect of the inflammatory cytokines on conjunctival goblet cell function remains largely unkn...

Immune profile of squamous metaplasia development in autoimmune regulator-deficient dry eye

PURPOSE Squamous metaplasia of the ocular surface epithelium in severe Sjögren syndrome (SS) dry eye has been implicated to be associated with chronic engagement of immune-mediated inflammation. While the detailed immunopathological mechanism underlying keratinization of the ocular muco-epithelium in this setting remains unclear, mice deficient in the autoimmune regulator gene (Aire) demonstrat...

Goblet Cells Contribute to Ocular Surface Immune Tolerance—Implications for Dry Eye Disease

Conjunctival goblet cell (GC) loss in dry eye is associated with ocular surface inflammation. This study investigated if conjunctival GCs contribute to ocular surface immune tolerance. Antigens applied to the ocular surface, imaged by confocal microscopy, passed into the conjunctival stroma through goblet cell associated passages (GAPs) in wild type C57BL/6 (WT), while ovalbumin (OVA) was retai...

Lipoxin A4 Counter-regulates Histamine-stimulated Glycoconjugate Secretion in Conjunctival Goblet Cells

Conjunctival goblet cells synthesize and secrete mucins which play an important role in protecting the ocular surface. Pro-resolution mediators, such as lipoxin A4 (LXA4), are produced during inflammation returning the tissue to homeostasis and are also produced in non-inflamed tissues. The purpose of this study was to determine the actions of LXA4 on cultured human conjunctival goblet cell muc...

Alteration of ocular surface mucins in MUC5AC-DTA transgenic mice

PURPOSE To investigate the compensation of secretory mucins with membranous mucins in mice with goblet cell deficiency. METHODS A transgenic mouse model in which conjunctival goblet cells were targeted was generated, and the expression of mucins was evaluated through the toxicity of diphtheria toxin A driven by a human mucin, MUC5AC, promoter. Immunohistochemical staining, in situ hybridizati...
